This study aimed to develop a rapid method for micropropagation of Melaleuca leucadendra. Nodal explants were washed and decontaminated with 0.1% HgCl2 for 16 min with the highest survival rate of 75.6%. The cultivation of nodal segments in basal WPM (Woody Plant Medium) supplemented with 30 g l-1 sucrose, 9.0 g l-1 agar, and 0.5 mg l-1 BAP resulted in the highest axillary shoot formation (3.0) after a month. The full-strenght WPM with 1.5 mg l-1 BAP and 0.1 mg l-1 TDZ proved effective for shoot multiplication (11.4 shoots), while the optimal for rooting was half-strength WPM with 1.0 mg l-1 NAA (100% rooting with 4.3 roots).

Key words: in vitro culture, rooting, shoot multiplication, WPM
