INDUCTION OF SOMATIC EMBRYOGENESIS IN RESPONSE TO THE APPLICATION OF CYTOKININS AND AUXINS DURING MATURE EMBRYO CULTURE OF PINUS NIGRA ARN

Abstract
Induction of somatic embryogenesis from mature Pinus nigra Arn. embryos were achieved after four transfers to different phytohormone containing media. Active proembryogenic cell proliferation was induced by cytokinins (6-benzyladenine, kinetin, zeatin) in mature cotyledon culture. These proembryogenic cells were used as the initials for somatic embryo formation in the presence of 2,4- dichlorphenoxyacetic acid plus kinetin, or zeatin. After four weeks, embryogenic calli were established. The culture of selected calli in the presence of 2,4- dichlorophenoxyacetic acid and kinetin, or zeatin facilitated embryogenic tissues formation and occasionally plantlet regeneration. Embryogenic processes were facilitated and accelerated by use of DCR mineral solution (Gupta and Durzan 1985). QLP (Quorin and Le Poivre 1977) media stimulated further somatic embryos and plantlet development
